上機檢測前,須用待測細胞制備質控樣本來設定流式細胞儀的熒光補償和設置十字門的范圍: (I) 未轉染GFP等標記的細胞 ① 空白管:陰性對照組細胞,沒有染色的細胞。不加Annexin V/APC,PI。用于調節電壓。 ② 單染管:有明顯凋亡的細胞,只加Annexin V/APC染色。用于調節補償。 ③ 單染管:有明顯凋亡的細胞,只加PI染色。用于調節補償 ④ 檢測管:待測的處理細胞,加Annexin V/APC,PI。用空白管和單陽管調節好電壓補償后,獲得所需要的流式數據。
(II) 轉染GFP細胞 ① 未轉染空白管:未轉染細胞,陰性對照組細胞,沒有染色的細胞。不加Annexin V/APC,PI。用于調節電壓。 ② 轉染GFP空白管:轉染GFP的對照組細胞,沒有染色的細胞。不加Annexin V/APC,PI。用于調補償 ③ 未轉染單染管:未轉染且有明顯凋亡的細胞,只加Annexin V/APC染色。用于調節補償。 ④ 未轉染單染管:未轉染且有明顯凋亡的細胞,只加PI染色。用于調節補償 ⑤ 檢測管:待測的處理細胞,加Annexin V/APC,PI。用空白管和單陽管調節好電壓補償后,獲得所需要的流式數據。
