CCM-300葉綠素含量測量儀是使用葉綠素熒光比率對葉綠素含量測量的一種新型儀器。與傳統的化學測量方法和吸收性測量方法相比,它可以對非常小的葉片以及難以測量樣品的葉綠素含量進行非破壞性測量,測量迅速可靠。
    一用途

    葉綠素含量測量儀廣泛應用于植物生理生態研究,特別是對傳統吸收法測量困難的小葉片或樣品的葉綠素含量進行測量,例如針葉、發育未*的水稻、生于巖石上的絲狀藻、地衣、草坪草、仙人掌、龍舌蘭屬植物、菠蘿、擬南芥、果實、苔蘚、葉莖、葉柄。

    二原理

    盡管實踐已經證明了吸收技術測量葉綠素含量的有效性,但該方法對樣品的要求較高,使其應用范圍受到限制。而樣品本身的葉綠素熒光則不受樣品厚度、均一性等的影響,可以指示葉綠素的含量。根據這個原理,Gitelson等比較了葉片的吸收和反射對葉綠素熒光的影響,發現在735nm和700nm處的比值與葉片的反射相關性最高,并且當樣品葉綠素含量在41mg·m-2到675mg·m-2時,F735/F700的比值與其葉綠素含量間存在著顯著的線性關系(R2≥0.95),與傳統的吸收性測量方法相比(測量范圍≤300mg·m-2),提高了葉綠素含量的線性關系范圍。

    根據Gitelson的研究結果,CCM-300使用具有15nm半峰寬的460nm二極管用于熒光激發,提高了紅色熒光波長,避開了大約685nm的發射熒光峰(葉綠素可重新吸收,降低測量精度)。儀器使用在735nm和700nm的葉綠素發射熒光的比率(F735/F700)進行葉綠素含量的測量,讀數為F735/F700的比值以及根據Gitelson方程計算的葉綠素含量。
