"作為后基因組時(shí)代表達(dá)克隆和蛋白質(zhì)純化技術(shù)zui重要的供應(yīng)商之一，德國IBA公司開發(fā)了一個(gè)稱為Strep-tag的通用技術(shù)平臺(tái)。這種快速、合算、多樣性地生產(chǎn)和使用重組蛋白的技術(shù)平臺(tái)是IBA與TU Munich的Arne Skerra博士教授密切合作開發(fā)而成。除了圍繞Strep-tag的大型產(chǎn)品組合外，IBA還*提供應(yīng)用該技術(shù)的蛋白質(zhì)生產(chǎn)定制服務(wù)。Strep-tag技術(shù)開發(fā)的原理是*的生物素（biotin）和抗生物素蛋白鏈菌素（streptavidin）之間的結(jié)合反應(yīng)。為了將這種牢固的相互作用用于蛋白質(zhì)純化用途，研究者們發(fā)現(xiàn)需要一個(gè)肽，該肽與重組蛋白質(zhì)融合后，能夠結(jié)合在streptavidin的生物素結(jié)合口袋，從而作為純化tag。zui后研究者們成功設(shè)計(jì)出一個(gè)只由8個(gè)氨基酸(Trp-Ser-His-Pro-Gln-Phe-Glu-Lys)構(gòu)成的短序列并稱之為Strep-tag II。為了優(yōu)化結(jié)合屬性，streptavidin也被設(shè)計(jì)成為Strep-Tactin。如此，一對優(yōu)化的結(jié)合伙伴被發(fā)現(xiàn)了：Strep-tag / Strep-Tactin系統(tǒng)現(xiàn)已成為zui被廣泛應(yīng)用的親和層析系統(tǒng)之一。Strep-Tactin與多種不同的基質(zhì)偶聯(lián)，使Strep-tag融合蛋白得以在生理?xiàng)l件下親和純化。與其他tag相比，這些溫和的純化參數(shù)能保存蛋白質(zhì)的生物活性，并僅經(jīng)一步層析后即可產(chǎn)出超過99%的純度。
